Title: New Approaches for Manufacturing Plasma Proteins
Abstract:
Cohn plasma fractionation process, developed by great scientist Edwin J. Cohn more than 80 years ago, is based on series of EtOH precipitation steps . The process is based on the differential solubility of albumin and other plasma proteins based on pH, ethanol concentration, temperature, ionic strength, and protein concentration.
The Cohn process derived proteins are very well characterised and proven to be very safe despite HIV and other potential contaminants which are these days well addressed.
Still, due to low processes yields, expensive equipment, huge size facility needs, facility flexibility, lengthy processes and use of the EtOH new approaches are sought.
This paper will address some of the bottlenecks of the Cohn fractionation and provide insight in some novel approaches which would allow for improved recoveries, small facility footprint, manufacturing flexibility, less EtOH usage and integral manufacturing. Novel in-process analytics to enable faster process development and better in-process control will be presented.
