Title: Monoliths as an Effective Tool in Vaccine Production – Showcases of Fast And Efficient Virus Purification
Abstract:
Maintaining the final product’s purity and safety is a critical challenge in biomanufacturing, especially when maximizing production efficiency, such as vaccine production. Modernizing existing production processes is an open challenge, as higher productivity should not compromise product safety.
Recombinant vaccines are often purified by chromatography due to its scalability, purity and high recovery rates. It also enables process intensification by introducing fewer, more efficient purification steps and reducing costs. Convective Interaction Media (CIM®) monoliths are proven tools for the purification of large biomolecules, such as viruses, virus-like particles, and nucleic acids, commonly used in vaccines, cancer immunotherapy, or gene therapy.
In this talk, three different cases will be presented. For Influenza and Newcastle disease virus purification, we developed one-step purification from clarified harvest to formulated product using CIMmultus® SO3 column. This approach achieved extreme column capacities, fast flow rates and high overall recovery (>80%), leading to higher productivity compared to traditional methods. The presentation will also cover the DoE optimization of lentivirus purification using CIM® monolithic 96-well plates, which increased the infectious lentivirus process recovery to approximately 60% with the CIM® QA column.
In all cases, monolith chromatography significantly enhanced overall productivity and produced high-purity products.
